Treesearch Research Infrastructure Seminar: Electron Microscopy seminar and Ultramicrotomy workshop

with Application Specialist Gareth Morgan from RMC Boeckeler and Dr Sofia Pettersson from Rowaco

Welcome to a Treesearch Electron Microscopy two-day Workshop on Ultramicrotomy with Application Specialist Gareth Morgan from RMC Boeckeler and Product Specialist Sofia Pettersson from Rowaco.

Ultramicrotomy is routinely used to produce thin sections and/or smooth surfaces for subsequent TEM, SEM and AFM imaging. The quality of the resulting sections is determined by many factors, including the material properties, sample pretreatment and cutting parameters. Knowing your sample is crucial to match the material properties with suitable cutting conditions.

This workshop will introduce some of the vast number of protocols that can be used to prepare materials of various kinds for sectioning and discuss the possibilities and limitations with the various techniques. Day one will mainly be focused on room temperature applications and day two will highlight ultramicrotomy under cryo conditions.

The theoretical sessions on both days will be live streamed (link will be provided to the registered). 

Please note that the available number of participants for the practical sessions is limited.

The workshop is free of charge for you who are associated to Treesearch or working at a Treesearch partner.

DAY ONE (MARCH 4TH):

Introduction to ultramicrotomy
Sample preparation protocols for materials sciences (general overview)
Advanced techniques for challenging samples

12:00 – 13:00 Lunch

13:00 – 16:30 Practical session (G. Morgan)
Demonstration of ultramicrotome
Room-temperature sectioning of TEM samples (embedded samples)
Room-temperature techniques for smooth surfaces
Hands-on ultramicrotomy


DAY TWO (MARCH 5TH):
09:00 – 12:00 Theoretical session (G. Morgan and S. Pettersson)
Introduction to cryo ultramicrotomy
Advanced cryo-techniques for soft/challenging materials

12:00 – 13:00 Lunch

13:00 – 16:30 Practical session (G. Morgan)
Demonstration of cryo-sectioning and lift-out techniques
Hands-on cryo-sectioning

Discussion and closing remarks

BIOGRAPHIES:

Gareth Morgan began his career in Electron Microscopy in 1971 as a Junior Technician in the Zoology Department of Kings College, University of London. Leaving Kings as Chief Technician and Laboratory Manager of the Multi Faculty EM Unit, he joined RMC in 1989 and is now a Senior Applications Scientist involved in teaching and training in the use of instrumentation and applied techniques.

Sofia Pettersson holds a Master of Science in Engineering and a PhD in Medicine from Linköping University. With a background in biomaterials, surface characterization and cell-material interactions, she has a keen interest in interdisciplinary research. Prior to joining Rowaco she worked as a materials engineer for polymers at a material testing and product qualification testing company. She is now the Product Specialist for EM sample preparation and microscopy at Rowaco.

The seminar is organized as a part of the Treesearch Electron Microscopy Series in a collaboration with RMC Boeckeler (Tucson, AZ, USA) and Rowaco AB (Linköping, Sweden).
